The problem is that you really can't tell that it is HTTP traffic unless
it is port 80. Port 80 is the only registered HTTP port.
If you are talking about transparent proxy for other protocols, then it
is a completely different issue and requires other software than Squid,
but in principle it is the same (many other protocols are however far
more complex to proxy than HTTP).
