We run two squid boxes here, 17000+ users and about 4000 workstations.
We enforce cacheing on some networks (saves bandwidth and also gives us
some extra security).

We peer with a number of nearby institutions and also use a national
system as parents. See:

http://wwwcache.ja.net/

For some more information (and yes, I'm also part of that project!)

We've found approximately a 40% saving (very rough guestimate) in our
outgoing bandwidth; as a whole in the UK the national service carries
roughly 50% of all inbound transatlantic web traffic for the JANET
network as a whole.

So to answer the questions in order:

Yes
Lots
Very

:)
