If you by dnsserver refer to "--disable-internal-dns" then no.

If you by dnsserver refer to running a caching DNS server nearby Squid
somewhere instead of using the ISP DNS server then yes. You want a DNS on
the local lan of Squid on the same side of any network bottleneck you
might have.
